Abstract

Human immunoglobulin G (IgG) in an antibody used for several therapeutic indications. As high concentration and purity are required, selective adsorption techniques, such as affinity chromatography, are recommended for recovery and purification purposes. Thus, this research project aims to evaluate human IgG adsorption in non-conventional affinity-based adsorbents synthetized of chitosan and alginate with Reactive Green, Reactive Blue and Cibacron Blue dyes immobilized as ligands. The adsorption will be evaluated under different experimental conditions, by varying the pH between 6.0 and 8.5 and the temperature between 4 and 37o C. The adsorption data modelling will be performed by using artificial neural networks and compared to classic non-linear methods for Langmuir, Langmuir-Freundlich and Temkin parameters adjusting. Adsorption thermodynamic parameters, such as Gibbs free energy (DG0), enthalpy (DH0) and entropy (DS0) will be estimated via artificial neural networks and van't Hoff and Gibbs-Helmholtz equations. Thus, it will possible to obtain the necessary data to define the optimum adsorption condition aiming to improve the use of this technique in downstream processing of human IgG. (AU)
