Abstract

Monoclonal antibodies (MAbs) are immunoglobulins secreted by a single clone of normal or tumoral B lymphocytes, or hybridomas, They have been widely employed in immunochemical and immunodiagnostic tests such as the detection and characterization of various molecules of interest in the clinical, research and biomedical applications which requires biologically active and high purity MAbs. Thus, this research project aims at the recovery and purification of anti-TNP MAbs from cell culture supernatant aiming its subsequent application in the composition of diagnostic kits. To this end, it will be studied purification strategies combining ammonium sulfate precipitation and affinity chromatography with immobilized dyes on epoxylated chitosan/alginate microspheres. In order to achieve this purpose, it is intended to seek a recovery and purification process in which the MAbs could be obtained with a purity exceeding 95% and at the same time not incurring losses in biological activity.
