Functional activity of mononuclear cells of colostrum of mothers with overweight and obesity and the role of immunomodulators agents: adiponectin, leptin and melatonin.

Abstract

Obesity is an important public health problem. Need special attention in critical phases as it is in pregnancy and infancy. There is evidence that maternal obesity promotes changes in the newborn metabolism that can lead to increased risk of the developing obesity and other diseases during adulthood. The breast milk is an important modulator of the metabolism and the child's immune system. Human milk has constituents in their composition with ability to influence the regulation of energy metabolism, such as adiponectin, leptin and melatonin. These bioactive substances also have activities on the functioning of immune cells. Obesity causes changes in the breast milk constituents, however it is not known how much obesity affects the action of mononuclear cells that are transmitted to the infant via colostrum. Therefore the objective of this study is evaluating the mechanisms involved in the activation of leukocytes of overweight and obesity women and the modulatory role of adiponectin, leptin and melatonin. The colostrum mononuclear cells will be separated by concentration gradient and are used in assays phagocytosis and oxidative burst, intracellular calcium and apoptosis. Will be added adiponectin, leptin and melatonin stimuli and analyzes will be performed by flow cytometry. Thus expected to generate specific knowledge about the influence of overweight in the function of mononuclear cells from human colostrum, which contributes to better care of maternal and child health. This is still a subject little described in the literature and can cause repercussions on the scene of Public Health, especially in the field of future obesity. (AU)
