Abstract

Endophytic fungi have shown promising sources for obtaining enzymes with biotechnological potential. Thus, during the course of this project will be obtained and purified strains of endophytic fungi associated with cassava leaves, Manihot esculenta Crantz. After obtaining the lines, it will be cultured in potato dextrose agar (PDA). The pure strains will be cultured using a specific medium for the production of laccase. For this, it will be used a culture medium composed of malt extract, Tween 80, copper sulfate and 2,5-xylidine. After growth, fungi will be separated and subjected to enzymatic extraction process to obtaining laccases. During this stage, the best extraction buffer is checked and also the presence of the enzyme in each previously isolated and cultured lineage. In parallel, studies related to formation of crosslinked in corneas and in hydrogels for the replacement of vitreous will be performed using laccase. Thus, the influence of the same the formation of cross-links will be verified, and determined of the optimal pH, reaction time, the enzyme concentration and temperature. For this, it being used a statistical planning. Physical, chemical, mechanical, thermal and surface tests will be conducted to evaluate the presence of cross-link in corneas and in hydrogels. Finally, the extracts that show the presence of laccases will be tested under conditions determined beforehand by performing a pre-purification of the enzyme of interest. (AU)
