Determination of the metabolomic profile of non-Hodgkin lymphoma for the identification of tumor biomarkers

Abstract

Non Hodgkin lymphoma prognostic has been achieved by the International Prognostic Index lately, which is obtained from physical characteristics, pathological observations and symptoms reported by the patient. Therefore, searching for prognostic and prediction tumor biomarkers for non Hodgkin lymphoma is crucial, aiming to preview more accurately and earlier the disease progress as well as the suitable treatment to each subject. The metabolomic profile of non Hodgkin lymphoma will be investigated by LC-MS/MS, GC-MS and CE-MS/MS in this project, so that prognostic and prediction biomarkers may be identified. Firstly, blood serum and urine samples from healthy subjects will be analyzed by LC-MS/MS, GC-MS and CE-MS/MS for optimization of analyses conditions. Sample treatment for LC-MS/MS and CE-MS/MS will be as simple as possible, so that analytes loss is circumvented. On the other hand, sample treatment for GC-MS will require more extensive biological sample manipulation in order to obtain volatile and or more thermally stable analytes. For this purpose, three procedures will be evaluated: conventional derivatization; volatile organic compounds analyses; on fiber solid phase microextraction derivatization. Concomitantly, blood serum and urine samples from non Hodgkin lymphoma subjects will be collected and stored before each chemotherapy cycle, and will be analyzed by the developed methods. After data treatment by multivariate statistics, the potential biomarkers will be validated and will possibly aid on the treatment to be used for each subject by a custom medicine approach. (AU)
