Abstract

This project is a continuation of the research line that has been supported by FAPESP (2013/22605-1; 2013/00976-8 ; 2013/24252-9 ; 2010/17534-0; 2010/16115-3; 2010/06395-9; 2006/56435-1; 2006/56759-1; 2003/01233-7. The objective is the systematic study of the structural and kinetic properties of the (Na,K)-ATPase in the gill tissue of different species of crabs and ATPase shrimps, aiming to establish a model for the adaptation of crustaceans to different salinity environments. The maintenance of osmolality of the hemolymph of crustaceans is attributed to the (Na,K)-ATPase and this adaptation mechanism suggests that the responses of crustaceans involve a rapid activation of ion transport that should be primary to any other physiological response of this animal.The objective of the present project is to evaluate comparatively the kinetic and molecular characteristics of the gill (Na,K)-ATPase of M. amazonicum populations inhabiting both coastal and continental areas, in order to access the elucidation of the physiological mechanisms of geographically separated entities and trying to clarify whether different populations of M. amazonicum found in different parts of the country are originated from a single species or they represent indeed different species. To achieve the objectives the following steps will be are performed : preparation of membrane-rich gill (Na,K)-ATPase from shrimps collected in different habitats; electrophoresis and Western blotting; determination of osmolality and ion concentrations in the hemolymph; mRNA expression of alpha subunit of (Na,K)-ATPase and V(H)-ATPase; FXYD2 effect on (Na,K)-ATPase activity.
