Abstract

Introduction: Polycystic Ovarian Syndrome (PCOS) is a metabolic disorder that affects around 10% of women of reproductive age. Insulin resistance and lower adiponectin serum levels are these disease key markers. The bone marrow adipose tissue (MAT) seems to be an important source of adiponectin production. Since that, we aim to see if PCOS and obesity's insulin resistance are associated with MAT expansion and inflammatory status, which leads to a decrease expression and secretion of adiponectin. AIM: To evaluate MAT expansion behavior, adipose tissue's adiponectin expression and serum levels of adiponectin in PCOS and obesity mice models. In addition, to quantify serum levels of bone formation and resorption, bone marrow inflammatory cytokines expression; trabecular and cortical bone mass. Methods: 40 female mice strain C57BL / 6J will be use. At 25 days of life, they are going to be allocated into 4 groups of 10 animals. Two groups will be treated with 60 days release dehydroepiandrosterone's (DHEA) pellet and divided into normal or high calorie diet. The others two will implant innocuous pellet and divided in normal and high calorie diet. The animals will be euthanized at 90 days of life and will have specific biological material selected and analyzed. Results: Insulin resistance will be measure by insulin tolerance test; evaluation of MAT expansion by histological and micro-computed tomography (microCT) analysis; assessment and quantification of the expression of adiponectin by Western blot analysis and AdipoQ gene by qRT-PCR. Serum adiponectin, osteocalcin, osteocalcin not carboxylated, telopeptide C-terminal (CTX1) and bone marrow TNF and RANKL expression are going to be measure by Elisa. Expression of adipocyte markers will be evaluated by qRT-PCR and volumetric bone mass (vDMO) by microCT. (AU)
