Effectiveness of preventive treatments in the erosion of dentin: in vitro and in situ study

Abstract

The aim of this study will be evaluate in vitro and in situ the effectiveness of different preventive treatments on dentin (with and without abrasion) against tooth erosion. For the in vitro phase, 120 slabs of root dentin will be obtained (4x3mm) from bovine incisors. Half of each specimen surface will be exposed to treatments/erosive challenges (liquid and gaseous hydrochloric acid). This phase of the study will be factorial (5x2), and the factors under evaluation will be Surface Treatment at 5 levels: no treatment (GC); epigallocatechin-3-gallate (EGCG); epigallocatechin-3-gallate + chitosan (EGCGq); 4% TiF4 varnish base; Er, Cr: YSGG (0.5W- 8.92 J / cm2) + 4% TiF4 varnish base and Erosion at 2 levels: HCLl (pH=1,2, 6x / day / 20s) and HClg (pH=1,2, 6x / day / 20) for 6 days. The abrasion will be performed with electric toothbrushes for 10s / 200gf with dentifrice. The analysis will be according to the volume loss, step, wear profile, surface roughness using confocal microscopy 3D laser (LEXT OLS4000, Olympus), chemical changes, nanoindentation and transverse microradiography. For in situ phase, 30 volunteers will be selected and divided into 2 groups: Control-healthy and GERD-diagnosed or suspected disease (with dental erosion). Ninety bovine root dentin (2x2x2) will be submitted to the oral environment and preventive treatments: GC, and the two groups that will present better performance/results in both periods (15 and 30 days). The following analysis will be performed: volume loss, step, wear profile, surface roughness using confocal laser scanning microscopy (LEXT OLS4000, Olympus). The data obtained will be analyzed using the same criteria of the study in vitro and submitted to ANOVA and Tukey test (p <0.05). (AU)
