Investigation of the effect of cell sheet of human dental pulp stem cells associated or not with biophotonics (photobiomodulation therapy and antimicrobial photodynamic therapy) on murine tissue regeneration

Abstract

The objective of this study will be to investigate the regeneration of bone lesions where the cell sheets (CSs) engineering will be applied associated with photobiomodulation therapy (PBMT) and antibacterial photodynamic therapy (aPDT) in rats presenting basic pathologies (Type 2 Diabetes mellitus) and in rats where the medication-related osteonecrosis of the jaws (MRONJ) will be induced by anti-resorptive drugs. The hypothesis of the study is that the association of cell sheet culture engineering with biophotonic therapies (PBMT and aPDT) will improve the regeneration of bone lesions in the mice bearing Type 2 Diabetes mellitus, as well as the regeneration of MRONJ lesions. The study will have 3 steps: one in vitro and the other two in vivo. In the first step, in vitro, CSs will be obtained to be applied in the treatment of bone lesions of the animals in the following in vivo steps. In vivo, two studies will be performed: first, MRONJ will be induced in 84 Wistar rats by using zolendronate, a potent antireabsorption medicatio, followed by upper molar extraction. Oral lesions will then be treated with application of CSs associated or not to PBMT and / or aPDT. In the second study, CSs, associated or not to PBMT, will be applied in 72 diabetic rats for treating standardized critical bone lesions in the parietal bone. The material obtained in both studies will be fixed and submitted to the analysis: computerized microtomography (MicroCT), histological (HE), histochemistry (TRAP- Tartrate Resistant Acid Phosphatase) and immunohistochemistry (cell tracking-BrDU and expression of markers involved in the osteogenesis process).
