Abstract

The characterization of the role of Z disc proteins by means of frontier technologies in biological images allows the understanding of cellular and tissue functions in homeostasis and cardiac diseases. By means of isolated myocytes, we have particularly advantageous tools to understand the functional impact of Z-disc protein dysregulation. Recent genetic and biochemical studies have highlighted the Calsarcin-1 (CS1) proteins found on the Z disk. CS1 is characterized as a negative regulator of the hypertrophic Calcineurin-NFAT pathway, mediated by mechanical stimuli and calcium. Consequently, specific forms of cardiopathies under mechanical stress indicate that the myocyte Z disk plays an important role in the transduction of mechanical events in biochemists. Thus the objective of this project is to study the role of the CS1 proteins of the Z disk for the recognition of the sensing machinery, and to understand the location and subcellular compartmentalization of the signaling pathways and the morphofunctional alterations, in experimental models of cardiopathies under mechanical stress making use of new frontier technologies in images to understand how the Z disk performs important functions in mechanotransduction.
