Use of Er:YAG laser in preventing caries in pits and fissures: an in vitro and in situ study

Abstract

The aim of this study will be evaluate the Er: YAG laser irradiation effectiveness at caries inhibition in pits and fissures using models of studies in vitro and in situ. Human third molars will be used in occlusal slabs of 3mmx3mmx3mm. The slabs of in vitro study will be treated with sub ablative energy of Er:YAG laser. The parameters of frequency and water flow refrigeration will be tested. The groups will be divided into 13 groups and sub groups: 12 experimental groups and sub groups and control group. The area of exposure of specimens will be defined and they will be cycled by DES and RE solutions per 10 days. For sub superficial micro hardness test the specimens will be sectioned in perpendicular way of fissure and this wall will be tested. The marks will be realized at the sub superficial area of deep wall and side wall of pits and fissures. For analyze at polarized light microscope the samples will be sectioned in a micrometer. The images will be picked up and digitalized for near irradiated surface analyzes and measurement the size of lesion. The surface and sub surface of samples will be analyzed in MEV.At in situ study the slab will be treated with Er:YAG laser and divide into 4 groups: control and 3 experimental groups. The experimental groups will be the three best parameters found at in vitro study. The samples will be adapted in intrabucal device that will be used for volunteers per 14 days. The volunteer will dropped the sucrose solution over the samples for cariogenic challenge. In the same way of in vitro study will be realized the micro hardness test, polarized light microscope analyzes and MEV analyzes. The data will be analyzed about distribution for the use of appropriate statistical test.
