Abstract

The immunological response profile to cancer may give important informations on antibodies induced by tumor’ proteins. Hence, a better understanding of humoral antigens associated to tumors that can be recognized as non-self by the immune system may help evaluate the tumor diagnosis and prognostic. Phage-display technology has been largely used to identify different proteins’ epitopes. It is based on the fusion of peptides or proteins to viral capsidium. In the present study, we will develop synthetic peptide libraries linked to a pIII bacteriphage M13 (PhD-C7C and PhD-12, Biolabs) using tissue and serum from 30 patients with differentiated thyroid tumors, 30 thyroid adenomas and 30 normal thyroid glands. The samples will be adsorved in microtitration plaques, submitted to biopanning for non-biding phages elimination e selection of the phages able to bind to antibodies. Bioinformatic analysis of the recombinant peptides and immunoenzymatic assays (ELISA, Dot-Blot and immunohistochemistry) with the obtained clones will validate potential diagnostic markers.
