Abstract

This project aims to study the biological activity of violacein on different strains of Staphylococcus aureus, an important pathogen for humans and other animals through techniques of proteomics, functional genomics, analysis of antimicrobial activity (in vitro and in vivo). Previous results have shown that this compound showed bactericidal effect on strains of S. aureus, including strains of S. aureus methicillin resistant (MRSA) and S. aureus with intermediate resistance to vancomycin (VISA) in vivo model. The in vitro activity of violacein on S. aureus will be evaluated using techniques of MIC and MBC, time kill curves, protein analysis in the presence and absence of violacein (SDS-PAGE, two-dimensional electrophoresis and mass spectrometry) analysis of cellular ultrastructure by transmission electron microscopy and differential gene expression methodologies using high-throughput sequencing. The results are confirmed by real time PCR. Still, the in vivo anti-staphylococcal activity will be evaluated in an animal model.
