Abstract

Aedes aegypti is one of the high world distribution mosquitoes and has medical importance because it is Dengue and Yellow fever vector. The increased number of disease cases transmitted by mosquitoes is due to the large number of larvae procreation sites and to the increase of mosquito's resistance to commercial insecticides. Protease inhibitors interfere in mosquito larvae digestive process inducing to a delay on their growth and development as well as significant mortality rates. In order to contribute in the development of new strategies in mosquito larvae control, we will use our experience in protease inhibitors and phage display system to construct a mutant inhibitor library exposed on M13 phage particles, using as a template molecule, a thrombin inhibitor produced by the mosquito Aedes aegypti, AaTI (Aedes aegypti thrombin inhibitor). This library will be used in specific digestion enzymes inhibitors selection from Ae. aegypti. The selected inhibitors will be produced in the yeast P. pastoris, purified, characterized and used in larval toxicity tests. (AU)
