Abstract

The fig (Ficus carica L.) is a fruit of great global importance, being among the top twenty fruits exported by Brazil, in this regard, research that can find solutions to improve the culture would bring enormous contribution to it. Thus, genetic improvement, with the use of mutagens, is an important line of research, being necessary to gather information about this species, especially in relation to its genetic variability, to spread projects and handling are carried out. Given the above, this study aims to determine the existence of variability due to epigenetic DNA methylation in selections tree between themselves and when compared to the main commercial cultivar, Roxo-de-Valinhos, using the MSAP analysis and subsequent Polymorphic DNA sequencing in order to standardize the regions of methylation, analyzing reads through bioinformatics tools, because the molecular analysis by RAPD and AFLP techniques, as seen in a previous study, showed no polymorphism among them. The experiment will be conducted at the Faculdade de Ciências Agrárias e Veterinárias - UNESP - Jaboticabal in partnership with the Faculdade de Medicina de Ribeirão Preto - USP and the Universidade de Ribeirão Preto - UNAERP. Analysis of the banding produced by each combination of primer used in the MSAP analysis will be given the parameter "1" for the presence of band and 0 for the lack of bandwidth, thus obtaining a binary matrix. The quality of the bases of the chromatograms and the arrangement of the sequences are analyzed using computer programs such as Phred and Phrap and the reads resulted from MASP technical will be mapped by software BSMAP to determine the position of methylation in CpG islands.
