Denture liners present excellent tissue tolerance. However, one of the main problems associated with its use is Candida spp. colonization, which is the rationale why attempts have been incorporated antifungal agents in these materials, for example undecylenic acid (UDA). The purpose of this study is to verify the action of UDA released from denture liners on C. albicans and C. glabrata biofilms. It is intended an in vitro simulation of UDA released by immersing denture liner samples (10 mm x 2 mm) in human whole saliva. The product released will be measured by chromatographic analysis (HPLC). In addition, susceptibility of planktonic cells will be determined by minimal inhibitory concentrations (MICs), minimal fungicidal concentrations and time-kill curves. To assess whether the AUD is effective at the concentration released from material, monospecie biofilms of reference strains and clinical isolates will be developed on the surface of denture liner samples. Then, they will be analyzed to cellular quantify, bioactivity, bioarchitecture and virulence factors. The measurement of cellular quantify will be performed by serial dilution. Bioactivity of the biofilms will be measured by the XTT reduction assay. Confocal microscopy will be used to evaluate bioarchitecture. Virulence will be assessed by measurement of hydrolytic enzymes produced on exposure to UDA. Results will be evaluated with statistical tests defined after exploratory analysis at significance level of 5%.
