Dextransucrase from Leuconostoc mesenteroides FT045B: dextran production with controled molecular weight, production of alfa-ascorbic acid and bromo-dextran compound

Abstract

Dextransucrase (EC 2.4.1.5) is an extracellular enzyme produced by several species of Leuconostoc and Streptococcus. When this enzyme is excreted in the culture medium in presence of sucrose, it is possible to produce dextran, by the sucrose broken in glucose and fructose, that should be used in the construction of the structure of dextran by transference to the receptor site. The objective of this work is in first place develop and standard a dextransucrase FT 045B purification methodology, then when we acquire a high enzyme activity, it should be possible to produce the compound ±-glicosídeo ascorbic acid, a stable and antioxidant compound, to be used as topic care against free radicals, and the purified enzyme also will be used in the weigh molar controlled dextran production, focused in the production of clinical dextran, applying central composite design, in which the variables will be: temperature, sucrose concentration and enzymatic activity, during dextran production. This work also have as an objective the development of bromo-dextran compound production metodology in order to substitute potassim or sodium bromide in epilepsy care, because this drug have significant side effects. It is important to note that Professor Dr. Antonio Carlos Guimarães de Almeida, with who we are in contact, the coordinator of graduation in Neuronal Bioengenery in the Universidade Federal de São João Del Rei (MG), see a great possibility in the use of the new compound (bromo dextran) and he is able to use this in new tests and research works about epilepsy care.
