Abstract

Currently, many immuno-inflammatory processes have been associated with coronary disease atherosclerosis, pointing to harmful effects on the activation of macrophages and a beneficial involvement of antibodies and their fragments directed against the low-density-lipoprotein (LDL) oxidized. With this comes the need for research methods obteção these products, in which biotechnology has been very utilizanda, primarily using genetic engineering, which uses microorganisms as a model for protein expression. In these processes there is the Pichia pastoris, a methylotrophic yeast easy to handle, has a strong inducing agent, is able to perform post-translational modifications, shows rapid growth and good yields. Thus, this project aims to study the production of fragments of anti-oxidized LDL from the cultivation of P. GM pastoris using alternative means of cultivation and different operating conditions. Initially we will study the influence of alternative sources of nitrogen. Later we will evaluate the influence of the variables (inoculum concentration, concentration of inducer, agitation, pH and temperature). And finally we will study the kinetics of production of antibody fragments in a bioreactor selecting the best conditions indicated by factorial designs. Obtaining this biotech product will be of great importance for the development of new biomedical research and deserve attention in prevention, treatment and diagnosis of cardiovascular diseases such as atherosclerosis.
