Abstract

Restless Legs Syndrome (RLS) is a neurological disorder that is characterized by the need to move the legs, associated with paresthesia, uncomfortable sensations, and motor restlessness. During the long-term treatment with dopamine was observed an unusual phenomenon called Augmentation, was considered a consequence of treatment. Objective: Identifying an animal model of augmentation undergoing pharmacological treatment with a dopamine agonist. Methods: Will be used 20 male Wistar rats (3 months). The animals will be distributed in the treatment group (TTO) - of 10 rats, and the control group (CTRL) - of 10 rats. Initially, the animals will perform an adaptation to the open field (10 minutes). After the adjustment period will be started the treatment, where animals receive the drug or saline daily, always at the same time, for 28 days. Will be held tests in an open field on days 1, 7, 14, 21, and 28 of treatment. Pramipexole (PPX - 0.1 mg/kg) and saline (0.1 mg/kg) will be injected into the peritoneal cavity 15 minutes before the start of the period in which animals will be evaluated in the open field. The treatment will take place around the same time. Will be determined the protein expression of Tyrosine Hydroxylase (TH) and D1 and D2 receptors by western blot.(AU)
