Abstract

Cancer is one of diseases mainly caused by genetic changes that give cells, unlimited capacity of proliferation, inability to enter apoptosis, metastasis and angiogenesis. These factors are often originated by an imbalance in the expression of certain groups of proteins. In many tumors the protein Bcl-2 related apoptosis is hiperexpressa. Thus, it is essential that procedures are performed to decrease the expression of this protein. To make this feasible, it is common to use RNAi. As this is easily degraded it is customary to combine it with vectors. In recent years, there was great commitment of the international scientific community for these vectors were nanoparticles. Among all nanovetores used mesoporous silica nanoparticles showed higher capacity of cellular penetration. Moreover, being porous, having ordered channels, large surface areas and high compatibility with drugs were tested as carriers of drugs and genes, and RNAi drugs and with great efficiency and success.Thus, we will test whether the mesoporous silica nanoparticles produced by Prof.. Edessa Dr. Martins de Souza Barros (CDTN-CNEN), are jointly capable of carrying Bcl-2 and RNAi decreasing the amount of tumor cells in vitro models. In addition, we evaluate the reduction in the expression of this important protein by Real Time PCR. Still, compare that with the decrease in mRNA expression of the alveolar cells of basal cell adenocarcinoma of human lung (A549), human lung carcinoma large cell (NCI-H460), human pancreatic carcinoma (PANC-1) and human glioblastoma (NG97), immortalized human keratinocyte (HaCaT) alter their pattern of apoptosis, angiogenesis, metabolism, and drug resistance by the methodology of PCR Array. So invest in this area is of paramount importance, since new scientific frontiers will be opened, raising the status of the course and introducing them to one of the most relevant and most prestigious international nanobiotechnology is to treat tumor.
