Abstract

KIT protein is important during embryogenesis, including gametogenesis, melanogeneis and hematopoiesis. Pathological activation of KIT protein due to mutations of c-kit gene may result in altered functions of the protein (gain) may result in cellular growth impairment and tumor development. This has been demonstrated in hematopoietic neoplasms, melanomas, gynecological tumors, thyroid and lung cancer, seminomas and gastrointestinal tumor. In salivary gland neoplasms, only a few reports have shown that alterations in KIT gene are present; however, these studies only report alterations in adenoid cystic carcinoma. In a previous study we detected that protein c-kit is involved in the process of salivary gland morphogenesis and benign neoplasms of these glands. Based on this evidence, our study will investigate the expression of proteins involved in the Kit signaling cascade as well as mutations and post transcriptional regulation of Kit gene in salivary gland tumors. To meet this goal, we plan to include 50 cases of pleomorphic adenoma and 50 cases of adenoid cystic carcinoma, and to use the techniques of confocal microscopy, pyrosequencing and micro-RNA analysis.
