Electrochemical sensor and UV-Vis spectroscopy for investigate the interaction of the pesticides chlorpyrifos, carbaryl and their metabolites with calf thymus ds-DNA

Abstract

Chlorpyrifos (organophosphate pesticide), carbaryl (carbamate pesticide), and their metabolites 3,5,6-trichloro-2-pyridinol (TCP) and 1-naphthol, respectively, can interact with the structure of DNA. These interactions, characterized by intercalations on the double helix, breaking of the single or double strands, and formation of adducts, can cause genetic mutations and carcinogenesis. Methodologies by using UV-Vis spectroscopy and electrochemical genosensors have been applied for the evaluation of toxic analyte - DNA interactions. Thus, in this project, the aim is to evaluate the possible interaction of chlorpyrifos, carbaryl, and their metabolites with ds-DNA immobilized on the pencil graphite electrode. Since the spectrophotometric analysis of the interaction of the chlorpyrifos and carbaryl with DNA indicates hypochromic and hyperchromic effects (Carolina Gomes da Rocha, Process 2010/07527-6), the possible interaction of the metabolites TCP and 1-naphthol with ds-DNA will be also monitored by UV-Vis spectroscopy. These studies will complement the results obtained by DNA electrochemical biosensor by using the boron-doped diamond electrode and UV-Vis spectroscopy (Processes Fapesp 2009/08161-8 and 2010/07527-6).
