Abstract

The objective of this study is to evaluate if human dendritic cells (DCs) produce PGE2 and LTB4 in response to high and low virulent strains of P.brasiliensis. In addition, the role of pattern recognizing receptors in this production, and the modulatory effect of these eicosanoids in DCs functions will be studied. DCs obtained from monocytes treatment with IL-4 and GM-CSF will be incubated with dectin-1, mannose receptors, TLR2 and DC-SIGN agonists, challenged with high and low virulent strains of P.brasiliensis and evaluated for PGE2 and LTB4 production. In an other set of experiments, DCS were treated with PGE2 and LTB4 inhibitors and evaluated for MHC class II, coestimulatory molecules (CD83, CD80, CD86 and CD40) chemokines receptors (CCR-5, CCR-7 and CXCR-4) expression, and TNF- alfa, IL-6, IL-10 and IL-12 production.
