Abstract

Somatic embryogenesis (SE), asexual propagation of in vitro embryos, is considered as one of the most promising methodology for the study of embryonic development in plants, as well as for elucidation of physiological and biochemical processes that are difficult to be determined in great size species. Additionally, this technique of in vitro cultivation enable the propagation of clonal mass of elite genotypes and germplasm conservation of tree species. Despite all this potential, the ES technique has proven recalcitrant to some species of tree as in the case of brazilian-pine (A. angustifolia). The study of biochemical and physiological stimuli needed for the development of zygotic embryo and its subsequent application in the SE, has proven to be one of the most promising alternatives to overcome the recalcitrance of tree species in the SE route. Among these stimuli, recent evidence has pointed to the salicylic acid (SA) as potential signaling molecule for proper in vitro embryo development. The objective of this project is to quantify endogenous levels of salicylic acid (SA) at different stages of zygotic embryogenesis, and assess their effect on the formation and development of somatic embryos of A. angustifolia. The results to be obtained, besides contribute to a better understanding of the physiological and biochemical involved during zygotic embryogenesis, will complement the studies being conducted in BIOCEL / IB / USP, for the optimization of systems of somatic embryogenesis in A. angustifolia.
