Abstract

Pacu fish has excellent features, such as fast growing, that promote the success of his intensive farming. Muscle growth in fish occurs by hypertrophy and/or hyperplasia involving satellite cells proliferation and differentiation; these processes are regulated by the expression of myogenic regulatory factors and myostatin. The aim of this study is to evaluate the influence of different temperatures of pacu rearing on MyoD, myogenin, myostatin and Pax-7 expression at different growth stages. Juveniles (1.5 g weight) will be distributed in tanks containing water and maintained at 25ºC, 30°C and at room temperature (six replicates for each group), under distinct recirculation systems, during the whole experiment. The samples collection will be performed at the beginning of the experiment and at 15, 30, 45 and 60 rearing days (n=24 animals for each group). The animals will be anesthetized, sacrificed and white muscle samples from dorsal region collected and frozen in liquid nitrogen for molecular and morphological analysis. Histological sections obtained in cryostat will be submitted to hematoxylin-eosin stain in order to perform muscle fibers morphology and morphometry analysis. The quantitative gene expression analysis of MyoD, myogenin and myostatin will be performed by Real Time Polymerase Chain Reaction after Reverse Transcription (RT-qPCR). Detection of MyoD, myogenin, myostatin and Pax-7 proteins and the count of marked nuclei will be analyzed by immunofluorescence technique. (AU)
