Genotyping of Giardia duodenalis: detection of mixed infections and genetic recombination in samples of human origin

Abstract

Giardia duodenalis is a protozoan of worldwide distribution responsible for causing enteric infections in a wide variety of mammals, including humans. It is considered a species complex, in which the isolates can be classified into seven distinct genetic groupings (assemblages A, B, C, D, E, F, G), but morphologically indistinguishable. The genetic groups of G. duodenalis have different spectra of hosts. Two phenomena have been frequent in molecular studies: (i) occurrence of heterogeneous gene sequences, characterized by the presence of polymorphic sites, when the genes are amplified and sequenced with primers no specific assemblage and (ii) divergence between loci when different assemblages are marked by distinct genetic markers used in the same sample. Allelic sequence heterozygosis, genetic recombination and mixed infections are the explanations for such events; however, the only way to evaluate the real participation of these phenomena in samples of G. duodenalis would be through the analysis of a single Giardia cyst. To date no work of this nature was performed. In view of this lack, this project's main objective is isolation of cysts of G. duodenalis using the technique of micro-manipulation and subsequent genotyping by real-time PCR technique. The results to be obtained should bring contribution to knowledge of genetic and taxonomic of G. duodenalis, as well as the epidemiological knowledge of giardiasis.
