Dynamics of blood and alveolar phagocytes in calves with mannheimiosis

Abstract

The bovine respiratory disease complex is a serious animal health problem. Among the diseases of the complex the mannheimiose bovine pneumonic (MBP) is the most prevalent bronchopneumonia and have the major economic impact. Studies to understand the disease have been done evaluating several changes, however assessment of the immune system during the course of the disease are not clarified. The objective of this study is to evaluate the dynamics of local defense cells in animals with systemic MBP using an experimental model of disease induction and also evaluate the effect of the drug in vitro, the treatment with the cellular function of blood phagocytes and macrophages in bronchoalveolar lavage and the evolution of infection and treatment by physical examination and inspection of indirect airways via bronchoscopy. For this, 12 healthy black and white Holstein calves with six months of age will be used and divided into two experimental groups. The animals will be submitted to intrabronchial inoculation of Mannheimia haemolytica serotype A1 and have their samples of bronchoalveolar lavage and blood collected at times distributed prior to inoculation and after the treatment. Samples collected will undergo testing to assess immune techniques using flow cytometry, quantification of cytokines, and cytological study. Blood cell counts and physical examination will be conducted to monitor the animals. Statistical analysis of data obtained is performed using the computerized statistical program MINITAB (1998). The medians and percentiles will be calculated the as a measure of central tendency for the different immunological tests of BAL and blood. The data we present parametric distribution will initially be evaluated by F test (analysis of variance), and when significant, the averages will be compared by the Tukey test (Sampaio, 1998). When the data present non-parametric distribution, the variables will be analyzed by non-parametric Mann-Whitney and Kruskal-Wallis test. (AU)
