Abstract

Vitamin D - 1,25(OH)2D3 conducts its endocrine actions by genomic and non-genomic mechanisms. Several organs, including liver, pancreas, muscle, fat tissue and bone, express the gene for vitamin D receptor. Leptin - which is a hormone produced by fat tissue that has important effects on energy homeostasis and control of satiety - and osteocalcin genes both are controlled by vitamin D receptor endocrine actions. The effects of obesity on bone mass of adolescents are not fully understood. Studies have shown that bone density gain, typical of adolescence, may be greater in obese adolescents, however others suggest lower bone mass in these individuals, after adjusting for height. Adipokines such as leptin seem to have strong participation in the regulation of bone metabolism in this population. Objective: This study aims to investigate the relationship between 25 hydroxyvitamin D blood concentrations and single nucleotide polymorphisms in the vitamin D receptor gene with osteocalcin and leptin concentrations in adolescents, and to explore metabolic parameters related to overweight and obesity in adolescents. Methods: This cross-sectional study will be conducted with 400 individuals from 14 to 19 years. Demographic and socio-economic data will be assessed, as well as anthropometric measures and food intake. Body composition will be evaluated by DEXA (Dual Energy X-ray absorptiometry) and blood collection (12 hours fasting) to determine the presence of single nucleotide polymorphisms in the gene of the vitamin D receptor will be carried out PCR reactions (polymerase chain reaction). Bioelectrical impedance and blood collection (after 12 hour-fasting) will be held to biochemical analysis of 25 hydroxyvitamin D, parathyroid hormone, glucose, insulin, carboxylated and uncarboxylated osteocalcin, leptin, adiponectin, lipid profile and creatinine. PCR (Polymerase Chain Reaction) will be used to determine the presence of single nucleotide polymorphisms in the vitamin D receptor gene. (AU)
