Evaluation of the effect of tooth movement on the development of diabetes-induced neuropathy in rats

Abstract

Diabetes mellitus is a metabolic disease characterized by an abnormal increase in blood glucose resulting from the absence or deficiency in insulin production. Among the complications of diabetes are the peripheral sensory neuropathies responsible for the development of central nervous system sensitization. A recent study showed that diabetes enhances the inflammatory process induced by orthodontic tooth movement by increasing the levels of inflammatory mediators, which are known to be directly related to primary afferent neuron hyperexcitability. Thus, the orthodontic tooth movement in decompensated diabetics might be hypothesized to cause neuropathy in the trigeminal system. The present study will include Wistar rats with diabetes induced by streptozocin. After orthodontic movement (0, 1, 3, 6 and 12 days), the trigeminal ganglion of each animal will be removed and submitted to biochemical analysis (Western Blot, RT-PCR and ELISA) to measure levels of glutamate, substance P and calcitonin gene-related peptide; and the expression of NK1, AMPA and NMDA receptors. As a control, will be used normoglycemic and diabetic animals with insulin replacement. The data will be assessed by analysis of variance (ANOVA) or T test as appropriate. Multiple comparisons will be made by the Tukey test. The F test will be used to compare the regression of time-response curve concerning diabetic rats, normoglycemic and diabetic with insulin replacement. Significance level for all tests will be established at p <0.05.
