Abstract

This research project aims at the discovery of potentially bioactive secondary metabolites against Leishmaniasis in inflammatory processes, such as anti-viral agents or against micro-organisms resistant to antibiotics. It will be held the chemical bioactivity of crude extracts from two invertebrates collected on the coast of Rio de Janeiro, selected based on their profile of biological activities and chemical profile by HPLC-UV-MS. Crude extracts of sea squirts, sponges and nudibranchs be prepared and sent to bioassays to detect leishmanicide action, anti-inflammatory, anti-viral and anti-microbial. Moreover, the same extracts are analyzed by LC-MS-PDA. Will select two of the extracts that have higher potential in biological assays, and also present chemical profile consistent with the presence of secondary metabolites. After fractionation of the extracts, isolation and purification of pure substances, they will be identified through the analysis of spectroscopic data. Isolated in sufficient quantity (> 4 mg), such substances will be evaluated in bioassays in which crude extracts were active, so if you know the profile of activity and, if possible, their mechanism of action. Thus, the design is to exploit marine sources for the discovery of compounds that might serve as models for drug development.
