Abstract

Cancer is a major cause of morbidity and mortality worldwide. Head and neck squamous cell carcinoma (HNSCC) represents the sixth most common type of cancer and, unfortunately, most cases are diagnosed in advanced stages, which limits the chances of cure in spite of therapeutic options. There is evidence that the initiation and progression of cancer depend on several components of the tumor microenvironment. The present study aims (a) to analyze the set of proteins and metabolites in the secretome of HNSCC cells subjected to hypoxia and radiation, (b) to investigate changes in morphology, proliferation rate, migration, invasion , and in the secretome fosfoproteome caused by paracrine factors that were induced by hypoxia and radiation in HNSCC cells, (c) to investigate, in immune cells and normal fibroblasts, changes in morphology, proliferation index, and in the secretome fosfoproteoma caused by paracrine factors that were induced by hypoxia and HNSCC radiation cells, (d) to analyze the effect of silencing (or over-expression) of genes modulated by paracrine factors triggered by hypoxia and radiation, proliferation, morphology, migration and invasiveness of tumor cells. The results may contribute to understanding the effect of paracrine factors released by neoplastic cells under conditions of stress on the stromal cells and in adjacent neoplastic cells. (AU)
