Abstract

Breast cancer is the most incident neoplasia among women. In Brazil this type of câncer is the leading cause of death among women. Breast tumor microenvironment is enriched by a variety of factors, which together promote the development and progression. The malignant transformation of mammary cell activity involves changes in different genes and proteins, which results in uncontrolled cell cycle and changes on cell differentiation. Sex hormones are among many factors that induce and promote carcinogenesis. Alterations in hormone secretion result in imbalance of tissue homeostasis, leading to several changes of the environment, which may favor the appearance of cancer. Recent studies show that there are variations in the levels of ADAMTS-1 in tumor tissues compared to normal tissues, but the exact role of this protease and the mechanism involved remains to be elucidated. Previous results from our group show that estrogen, progesterone and testosterone are able to inhibit or promote the expression of mRNA ADAMTS-1 in normal and tumoral breast cells. In this project, we are going to study the effect of these hormones alone or in combination, and at different concentrations to evaluate the protein levels and subcellular distribution of ADAMTS-1.
