Abstract

The aim of this project is through a fed- batch fermentation, with the use of pH ( pH-stat) increase the efficiency of fermentation and avoid formation of plaster during production of lactic acid. For the production of L(+) lactic acid, by Lactobacillus rhamnosus B103 in batch will be use the optimized medium( 110g/L lactose, 90 ml/L corn step liquor, 2ml/L Tween 80 and 0,15 g/L manganese sulfate). Will be made five different types of fed- batch: simple batch, fed- batch with a constant flow, fed- batch by pulse, exponential fed-batch and fed-batch by pH stat. In all the experiments, samples will be withdrawn, each 6 hours, to kinetic evaluation of lactic acid fermentation. After determination of the best fed-batch process, will be made one fermentation using the best parameter and with the fermentation broth different purification methodologies will be evaluated: resin ion exchange, nanofiltration and conventional method.
