Abstract

The chronic consumption of ethanol results in significant changes in cardiac and circulatory functions, appearing as an important risk factor in developing cardiovascular diseases such as hypertension. In models of chronic consumption of ethanol was observed that the initial step for the ethanol-associated cardiovascular dysfunction involves the formation of reactive oxygen species (ROS), this process being mediated by the enzyme NAD (P) H oxidase. Angiotensin II (ANG II) activates NAD (P) H oxidase in the cardiovascular system and chronic consumption of ethanol induces activation of the renin angiotensin system (RAS). The quest for understanding the molecular mechanisms involved in cardiovascular effects of ethanol led to the study of the activation of MAPKs (mitogen-activated protein kinase) by ethanol since these proteins play an important role in intracellular signaling. The MAPKs are activated by different stimuli such as ROS, and ANG II. The hypothesis of this study is that the chronic consumption of ethanol stimulates the RAS which in turn induces the production of ROS (via activation of AT1 receptors) and activation of MAPKs. This process would lead to alterations in vascular reactivity and bioavailability of the NO as well as increased blood pressure. (AU)
