Abstract

Dendritic cells (DCs) are powerful antigen-presenting cells and, since the possibility of their in vitro generation from monocytes was described, many DC-based immunotherapy protocols have been developed, including for cancer. The loading of DCs with tumor antigens can be accomplished in different ways. One method that has been widely used is the fusion of dendritic cells with tumor cells. The defects that are found in dendritic cells derived from blood precursors of cancer patients led to the development of protocols using allogeneic DCs from healthy donors. The use of tumor cell lines or mRNA from tumor cells of the patient as a source of tumor antigens are strategies that aim to reduce the tumor mass required for treatment. Vaccination with DCs transfected with total tumor mRNA, however, have shown less clinical results than expected, probably due to the expression, by dendritic cells, of tumor inhibitory proteins, as a result of the use of total tumor mRNA. The aim of this project is to evaluate a different protocol of loading DCs with tumor antigens from patients: the fusion of stable tumor cell lines transfected with total mRNA from other tumor cell lines (as a model for the use of mRNA obtained from patients' tumors).
