Effects on lipid metabolism and plasmatic and tissue fatty acid composition in animals treated with HESPEREDIN before and after enzymatic hydrolysis

Abstract

Hesperidin, a glycosylated flavonoid found in abundance in oranges, has anti-oxidant, anti-inflammatory, immunomodulatory and other properties. Recent studies show the main limitation to the bio-availability of glycosylated flavonoids to be their relatively poor intestinal absorption. Enzymatic hydrolysis with total or partial liberation of the glycosyl fraction is capable of generating simpler compounds more readily absorbed. This study aims to make a comparative evaluation of the effects on lipid metabolism and the composition of plasmatic and cellular fatty acids in rats treated with hesperidin before and after enzymatic hydrolysis with hesperidinase. For the in vivo tests hesperidin (1mmol eq hesperidin/Kg animal weight), before and after enzymatic treatment, will be administered orally by gavage to male Wistar rats for 30 days. After blood sampling, plasma, erythrocyte and tissue (liver, skin, brain and lungs) lipids will be extracted using chloroform and methanol. Fatty acid composition will be analyzed by gas chromatography. The composition of the fatty acids will also be analyzed after separation of the phospholipids by solid phase extraction chromatography. Also the effect of the enzymatic treatment on antioxidant activity and on lipid metabolism will be assessed by quantifying malondialdehyde ( tio-barbituric acid-reactive substances -TBARS), total lipids and plasmatic lipids. The proposal is designed to contribute to understanding the antioxidant action of hesperedin and its hydrolyzed derivatives and to elucidate those effects in the light of this flavonoid's structure (AU)
