Evaluation of cytotoxicity and genotoxicity of desensitizing toothpastes on human pulp fibroblasts

Abstract

The purpose of this study is to evaluate in vitro the possible cytotoxic and genotoxic effects of toothpaste on human pulp fibroblasts (HPF-1). It will be established four groups: Group I- desensitizing toothpaste (Colgate Sensitive Pro-alívio, Colgate-Palmolive), Group II-desensitizing toothpaste (Aquafresh Sensitive, Aquafresh®), Group III- desensitizing toothpaste (Oral B Pro Sensitive, Oral-B®)and Group IV- control (Colgate Total 12® Professional Clean, Colgate-Palmolive). The cells will be exposed to toothpaste for 24 h, and cell survival will be measured using the MTT assay in a spectrophotometer (570 nm). Genotoxicity is determined by the formation of micronuclei in cells after 24 h of exposure to toothpaste and Ethyl methanesulfonate (positive control). The differences between the rate of cell survival and a number of micronuclei will be statistically analyzed by Mann-Whitney-U (p <0.05).(AU)
