Interaction of bioactive technologies to dentin on the obliterative potential of exposed dentin tubules by erosion

Abstract

Among the clinical events that affect the tooth structure, the most challenging ones rely on dentin. Due to the morphological and chemical structure and the relationship with the pulp tissue, the exposure of the dentin tissue to some stimuli can generate painful responses, a clinical condition called dentin hypersensitivity. To reduce the effects of this condition, strategies based on different mechanisms of action result in some alternatives in which toothpastes and gels are the main vehicles. Among them, bioactive technologies have shown promissory, due to their compositions and positive interactions with biological tissue. Therefore, it is relevant to understand the interaction of available desensitizing products, whether for home or professional use with the dentin. The objective of this in vitro, parallel and blind study is to comparatively analyze products based on different bioactive technologies on the obliterative potential of exposed dentin tubules by erosion. The analysis will involve two factors: material, in 4 levels (control- Colgate total 12, Elmex, Biomin and 5% SPRG paste), and condition, subdivided in 2 levels (control- artificial saliva and eroded in orange juice). The main response variables to be studied are wettability, surface microhardness and qualitative surface analysis by scanning electron microscopy and laser confocal scanning microscopy. 48 caries-free human third molars will be used, which will be stored in a 0.1% thymol solution, pH 7.0. The crowns will be sectioned using a diamond disc under cooling with deionized water. To measure the wettability each dentin surface will be tested by calculating the contact angle. For the evaluation of surface microhardness, a MicroMet 6040-Buehler microhardness meter will be used for 3 Knoop readings per specimen (10KgF/10s). Microscopy images will be qualitatively assessed. Statistical analyzes will be processed and in case of normal distribution, the Analysis of Variance tests and analysis of multiple comparisons will be applied by 2-way ANOVA and Tukey tests (p<0.05). Then, half of the specimens will be subjected to scanning electron microscopy.
