Pregnancy rate, inflamatory response and uterine blood flow affected by the effect of Pentoxifylline and sêmen quality assessed by fluorecent probes

Abstract

The aim of this experiment is to evaluate the effect of the semen extender containing Pentoxifylline and effects of the artificial insemination with different quality semen on the uterine blood flow and fertility rate, as well as the effects of the addition of pentoxifylline on the semen extender on sperm parameters and on the equine uterine clearance process. Semen samples of only one stallion will be assessed using a protocol of fluorescent probes combination: PI, FITC-PSA e JC-1 and clustered according to percentage of cells with plasmatic membrane integrity, intact acrossome and high mitochondrial membrane potencial (PIAIC). Considering semen with high quality (40-60% PIAIC spermatozoa) and low quality (10-30% PIAIC spermatozoa). For this experiment 24 mares will be selected and distributed in 6 groups. On the first cycle, 3 groups will be used: Group C (control): they won´t be inseminated, but submitted on artificial insemination mimic technique (n=8), Group D: uterine infusion only with semen extender without Pentoxifylline (Botu-sêmen®) (n=8) and Group DP: uterine infusion only with semen extender containing Pentoxifylline (Botu-turbo®) (n=8). On the second cycle, 3 other groups will be used: Group SS: artificial insemination with semen of high quality plus semen extender without Pentoxifylline (Botu-sêmen®) (n=8), Group SP: artificial insemination with semen of high quality plus semen extender containing Pentoxifylline (Botu-turbo®) (n=8) and Group B: artificial insemination with semen of low quality without pentoxifylline(Botu-sêmen®) (n=8). Uterine blood flow will be measured by transrectal color Doppler sonography spectral mode. The uterine artery will be scanned on 7 pre-determined periods: immediately before ovulation induction (follicle e 35mm) (T-30), immediately before artificial insemination (T0), 2 (T2), 6 (T6), 12 (T12), 24 (T24) and 48 hours (T48) after artificial insemination. Twelve hours after artificial insemination endometrial cytology will be done. Pregnancy rate will be assessed by transrectal sonography mode B on day 14 and 30 after artificial insemination. For data analyses SAS program, 9.2 version will be used.
