Adjuvant therapy with sodium alendronate for treatment of experimental periodontal disease in rats

Abstract

The aim of this study will be to evaluate the wound healing of periodontal tissues and the effects of mechanical treatment of scaling and root planing (SRP) in animals with induced periodontal disease, treated or not with locally alendronate sodium as adjuvant therapy. Induction of periodontal disease (PD) is performed by ligature placed around the first molars 90 of male rats. After 7 days of placed ligature, the animals will be divided into 3 groups of 30 animals each with the following treatments: Group F - Maintenance ligation throughout the experimental period; RAR Group: - Removal of ligation followed by SRP and irrigation with 1 ml of saline solution, Group AL: - Removal of ligation followed by SRP and irrigation with 1 mL of alendronate sodium (10-5M). Ten animals from each experimental group will be sacrificed on days 7, 15, and 30 days after treatment of periodontal disease. To analyze the results will be obtained semi-serial sections with a thickness of 4 µm for histomorphometric analysis and immunohistochemistry (CASPASE-3, OPG, and RANKL presence of TRAP-positive cells). The sections will be stained with hematoxylin and eosin and by Masson's trichrome technique. The data collected will be evaluated statistically with a significance level of 5%. (AU)
