Abstract

The generic delimitation of the subtribe Spiranthinae has been controversial since the first classification systems in Orchidaceae, but only recently studies based on phylogenetic relationships have clarified the position and boundaries of genera and groups within the subtribe, in particular the work of Salazar et al. (2003). For relations between the genera, the data of the nucleotide sequences of this study allowed the identification of major clades in Spiranthinae: "Stenorrhynchos clade", "Pelexia alliance", the pair Eurystyles/Lankesterella, and "Spiranthes clade." The cytotaxonomy, in turn, is the study of structural and numerical chromosome and its taxonomic use. The chromosome number is the most widely known cytotaxonomic parameters. This project will be carried out molecular phylogenetic analyzes to explain the phylogenetic relationships of the internal "Stenorrhynchos clade" and clarify the boundaries of genera therein. Furthermore, cytological data will be analyzed to know the chromosome numbers of species and describe the ideograms of the same clade. For the molecular phylogenetic analyzes will be used sequences of ITS (nuclear DNA), matK-trnK and trnL-trnF (plastid DNA), which will be analyzed by the methods of maximum parsimony, likelihood and Bayesian. Cladograms will be obtained separately for each marker as well as the consensus tree. For cytotaxomonic analysis will be collected root tips (an area where is found the root meristem) that will be pre-treated with a solution of 8-hydroxyquinoline 0.002M (8a Hq) and subsequently crushed on lamina, stained with Giemsa and observed on microscope for chromosome counting. It is worthy of exception that the phylogenetic analysis of the group will be an important contribution to establishing a solid basis for further taxonomic revisions of genera analyzed, and chromosomal studies are virtually nonexistent in the taxa of this clade.
