Abstract

Cyclodextrin glucanotransferase (CGTase, EC 2.4.1.19) is an enzyme able to forming cyclodextrin as the final product with starch as substrate. It is an enzyme that has gained industrial interest due to the increasing application of cyclodextrins (CDs) in the food and pharmaceutical industries. Two alkalophilic bacteria, Bacillus sp subgroup alcalophilus E16 and Paenibacillus campinasensis H69-3 were isolated from soil sample and selected because it has good production of CGTase. The forms of production and characterization of these enzymes have been studied and both showed potential for industrial application. An operation that limits the use of cyclodextrins is their obtaining from dextrinized starch by CGTase. Generally the yield is very low due to ring opening by the action of the enzyme itself and / or by changing the cavity encapsulation that occur during the drying process. Thus, this proposal aims to study the production of cyclodextrin by the action of CGTase from Bacillus sp subgroup alcalophilus E16 and Paenibacillus campinasensis H69-3 and the subsequent obtaining of CDs is the technique of spray drying (spray drying). Are measured process variables and the use of possible adjuvants necessary to maintain the stability of CDs. The CDs retrieved by spray dryer (spray dryer) will be analyzed compared to the drying process in an oven. Finally, the characterization and quantification of CDs (±, ², ³) will be performed by HPLC and high performance liquid chromatography with Dionex ion detector.
