Abstract

Germ cells (GC) are the precursors of the gametes. During their embryonic development, the GC undergo a wide epigenetic reprogramming, when DNA methylation is mostly erased. At this moment, the GC temporarily present pluripotency characteristics and can be transformed into stem cells in vitro. This capacity have been studied in mice, but not in rats. Our group has been showing that rat germ cell development is different from mouse and show some similarities with humans, what make the rat a good model to study germ cell development. One difference between mice and rats is the difficulties to get stem cells from the rat PGC, what has been well described in mice. Studies form our group have been suggesting that this can be due to the differences in the timing of the epigenetic reprogramming in these two species. Thus, the aim of this study is to change the period of epigenetic reprogramming in the rat PGC to extend the period when these cells show pluripotency characteristics. Pregnant Wistar rats will receive 5-Aza-deoxicitidina (5-Aza, 2mg/Kg) at 12dpc. The epigenetic reprogramming and Ssea1 expression will be investigated. This study can provide data to improve the derivation of pluripotent stem cells from rat PGC. (AU)
