The influence of gut serotonin inhibition on the pathogenesis of periodontal disease in rats: histological analysis

Abstract

Although biofilm is considered the main etiologic agent of periodontal disease (PD), tissue damage is partially given by localhost response activation. The hypothesis of gut serotonin (GS) inhibition influencing host response in PD is based on literature reports about the anabolic role of LP533401 into bone. LP533401 is an inhibitor of the tryptophan hydroxylase (Tph1) enzyme, important for gut serotonin production by enterochromaffin cells. The aim of this study is to evaluate the effect of GS inhibition during PD pathogenesis. Thirty male rats will be divided into three groups according to with PD ligature induction around lower first molars and treatment: (1) Treated Group: PD induction and LP533401 (25mg/Kg/day) by gavage; (2) Control Group without PD and (3) Control Group with PD, both treated with vehicle. All animals will be treated for 28 days (beginning 1h before ligature), when we will collect 3 ml of blood, and then animals will be sacrificed. Left mandibles will be decalcified, stained with Hematoxylin-Eosin and Picrossirius Red to analyze the alveolar bone loss and amount of collagen in the gingiva. Assessment of serum serotonin will be performed by ELISA. Quantitative results will be submitted to exploratory analysis to determine the appropriate statistical test that will be applied. (AU)
