The influence of the parasympathetic neuromodulation on induced periodontitis and on tibia micro-architecture in mice: macroscopic and histologic analysis

Abstract

Periodontal disease (PD) is a chronic oral inflammatory disease that begins with the accumulation of bacterial biofilm on the tooth surface, being the main cause of tooth loss in adults. Bone remodeling is a process regulated by several local and systemic factors and whose function is to maintain the bone mass through the formation by osteoblasts and resorption by osteoclasts. Recently, it has been shown that the systemic regulation of bone remodeling also involves two branches of the Autonomic Nervous System (ANS): adrenergic and parasympathetic or cholinergic. The objective of this study is to verify, by administering an acetylcholinesterase inhibitor, the effect of parasympathetic neuromodulation on induced PD and on the micro-architecture of tibia in mice. We will use thirty mice, divided into three groups: (1) Donepezil (D): induced PD in ten animals treated with 2 mg / kg / day Donepezil; (2) Ligature (L): ten animals in which PD will be induced; and (3) Control (C, n = 10). PD induction will be held by the insertion of cotton thread around the first mandibular molars and the treatment will be performed by gavage for 6 weeks. After sacrifice, the right hemimandibles will be submitted to macroscopic analysis for assessment of alveolar bone loss, and the tibia will be subjected to histological and histometric analysis. The data will be submitted to the most convenient statistical analysis.
