Abstract

This Doctorate project aims to characterize two regulatory systems from Caulobacter crescentus, FEC2 and FEC4. These systems comprise a sigma factor, an anti-sigma factor and a TonB-dependent receptor, and they are described in other bacteria as involved in the uptake of organic iron chelators (siderophores). Promoter expression assays and growth curves of mutant strains were performed in the presence of some iron sources, but all inorganic.Since the FEC systems from other bacteria are involved in the uptake of siderophores, it is interesting to evaluate the response of the FEC2 and FEC4 systems from C. crescentus to these molecules. In this subproject, we propose to study this through siderophore utilization assays and promoter expression assays in the presence of these compounds. The chosen laboratory offers a great variety of siderophores to test, allowing a rapid screening of the molecular signals to which the FEC systems of interest respond. This internship should be performed in July 2013, in a period of four to eight weeks. (AU)
