Abstract

Dependence on fossil fuel sources as well as concern for the environment has generated great interest worldwide in the development of other energy sources. In this context, the use of ethanol as fuel is advantageous because it is an inexhaustible source of energy, and is considered a low environmental impact. Currently, Brazil is the second largest producer of bioethanol which is produced from sugarcane juice fermentation. However, studies suggest that if sugarcane bagasse and straw were used, Brazil could double the production. Besides, the use of this biomass presents the challenge of the degradation of the plant cell wall structure in simple sugars which could be fermented. For this goal, many types of hydrolytic enzymes are needed. These enzymes are naturally produced by a variety of microorganisms, among which the fungus Trichoderma reesei, that is able to produce and to secrete them in large quantities. Although many studies have been carried out with this organism, the molecular mechanisms responsible for the degradation of these substrates are not clear. Thus, this work aims to analyze the global gene expression of T. reesei grown on bagasse and culm as exclusive sources of carbon by high-throughput sequencing of RNA technology (RNA-Seq). The goal is to understand how this fungus behaves in the degradation of lignocellulosic substrates, i.e., which genes and / or pathways are modulated in the presence of these substrates as carbon source. All resulting knowledge will contribute to the development of new enzymatic cocktails that can enable the large-scale production of second-generation ethanol. (AU)
