Abstract

Streptococcus pneumoniae is a common respiratory pathogen worldwide. The pneumococcal 7-valent conjugate (PCV7, including serotypes 4, 9V, 6B, 14, 18C, 19F and 23F) was licensed in the U.S. and Europe, showing a significant impact in reducing invasive disease caused by vaccine serotypes of pneumococcus. In Brazil, the National Immunization Program in 2010 included the 10-valent conjugate vaccine (PCV10, PCV7 serotypes plus serotypes 1, 5 and 7F). With the introduction of conjugate vaccines, several countries have reported an increase in non-vaccine serotypes in patients and as a major cause of invasive pneumococcal disease, particularly serotype 19A, considered of great concern because of its association with antimicrobial resistance and its high potential invasion. Molecular typing contributes to the knowledge of new clones or strains and their resistance movement in a particular region. In Brazil there is no information on the genetic lineage of the S. pneumoniae serotype 19A in the period before and after the introduction of pneumococcal vaccine PCV10. This study aims to evaluate the impact of the introduction of PCV10 vaccine in phenotypic and molecular characteristics of Streptococcus pneumoniae of invasive strains and carriers of serotype 19A in pre and post vaccination. Genetic characterization of pneumococcal strains will be performed by Pulsed Field Gel Electrophoresis (PFGE) and Multi Locus Sequencing Type (MLST) in strains belonging to serotype 19A strains selected from the database of the Center for Meningitis, Pneumonia and Pneumococcal Infections (NMPI), Centre of Bacteriology Instituto Adolfo Lutz, from January 2005 to December 2012.
