Abstract

In vitro production (IVP) in cattle is an important tool for animal reproduction. Despite its commercial and research applications, the results are considered unsatisfying. Sperm factors could reflect on embryo development during IVP. The correlation of seminal quality evaluation methods with fertility prediction has been strongly studied. However, the relation between fertility and seminal quality remains unpredictable, even with the advances in seminal analysis with the employment of specific probes and flow cytometers. These techniques allow analysis of a greater number of sperm cells and seminal parameters simultaneously. The goal of this work is to assess the effects of sperm parameters, such as plasmatic membrane integrity, intact acrosome, high potential of mitochondrial membrane, chromatin structure resistance and sperm DNA integrity, on IVP rates and embryo quality. In this study the effects of these parameters will be evaluated considering the cleavage time and rate, blastocyst rate and the analysis of expression profile of blastocysts, using as target genes some ones that show in literature correlated to embryo quality. This study proposes to clarify the effect and influence of each parameter on the fecundation process and early embryo development. IVP is not only an experimental model, it has economic relevance. The elucidation of the sperm effects could help on the choice of bulls or semen samples for breeding programs using reproductive technologies.
