Effects of 7-hydroxycoumarin on hematopoietic response of Listeria monocytogenes infected mice

Abstract

The increasing search for pharmaceutical drugs has been growing interest in coumarins, since these substances present relevant pharmacological activities, with no toxicity to mammals. Umbelliferone and 7-hydroxycoumarin (7-HC) is a natural product of the coumarin family, and in this study was extracted from the Typha dominguensis. According to reported pharmacological actions of 7-HC, especially in relation to its immunomodulatory potential, in this work we propose to study its effects on the hematopoietic response of Listeria monocytogenes infected mice. Studies have shown listeriosis as an effective experimental model to study the influence of several toxic and natural compounds in the immune response involved in host defense against LM, with emphasis on the ability to produce hematopoietic tissue, and mobilize phagocytes. During the initial phase of infection, occurs a decrease in the number of macrophages and granulocytes progenitor cells (CFU-GM) in the bone marrow and consequent migration to the spleen and extramedullary hematopoiesis, leading to splenomegaly and increased serum colony-stimulating activity (CSFs). In this line, we will use the Listeriosis model to determine if 7-hydroxycoumarin can modulate hematopoietic response of mice preventing the myelosuppression induced by infection. (AU)
